You can use Paint and set the duration to single frame. It records every stroke as a new 'brush' in the effect. I sometimes used that for creating guides for animation and then deleting them when I've got what I want. You can fake onion skinning, but the additional steps are too much. Instead opt for the more traditional animators 'flip' and store 2-3 previous frames in snapshots on F5-F8 (shift F5-F8 to store).

You can also do each frame as a layer in AI and when you import the AI file into AE as a Composition and sequence the layers everything is already done for you.

I did exactly this but I found it was kind of a weird way of doing things.
